ABOUT THIS TOOL

Compute average Ct, ΔCt, ΔΔCt and the 2^-ΔΔCt relative expression fold change from control and treatment data.

Important: Descriptive, group-level only. The calculator separately averages each Ct list and does NOT match target/reference by biological sample. For biological replication, compute each sample’s paired ΔCt before estimating variation and doing inferential tests. The 2^-ΔΔCt method assumes approximately equal, near-100% amplification efficiencies.

How it works

Method: ΔCt = Ct(target) − Ct(reference). ΔΔCt = ΔCt(treated) − ΔCt(control). Fold change = 2^(−ΔΔCt).

Example: If the control ΔCt is 4 and treated ΔCt is 2, then ΔΔCt = −2 and relative expression is 4-fold.
